ENCORE Priming Physiological Analyses Pipeline

This pipeline included the planned physiological assays to be completed for this project.

Pipeline Diagram

Diploria labyrinthiformis and Madracis decactis fragments kept in the -80°C freezer.

Protocols:

1. Airbrushing
2. Total Protein
3. Chlorophyll a & c
4. Symbiont counts
5. Carbohydrates
6. Ash-free dry weight
7. Total Lipid Content
8. Surface Area Wax-dipping

Once airbrushed, aliquot the following for each protocol:

• Total protein: 1mL
• Chlorophyll a & c: 1mL
• Carbohydrates: 1mL
• Symbiont counts: 500μL
• Total Lipid Content: 1mL
• Ash-free dry weight: 5mL
